I am going to replace my old piston (95 hours) by a Vertex cast piston.
In order to break up this piston I am thinking to do 3 heat cycles.

1) On the stand 5 minutes from idle to 1/4 then let it cool down
2) Another on the stand 5 minutes from idle to 1/2 throttle and let the engine cool down again
3) Riding around with a small load at 3 and 4 gear slowly for 10 minutes and then riding normally but without let the engine cool down.

Is it acceptable this procedure for a cast piston ?

Sounds good enough. Vertex pistons have a moly coating on the skirt to help during the break-in period.

Forged pistons require heat cycles. Cast pistons generally maintain size and shape during the first few operations better than forged pistons simply because that is the nature of a cast piston. It is a cast where as a forged piston is molded to shape while hot. This creates internal stresses in the forged piston that can only be released by heating and cooling several times. However it never hurts to play it safe.
