note: On any two stroke I have ever seen twisting the throttle does NOT "prime" the carburetor. Four strokes usually have accelerator pumps, two strokes do not.
A starting point that should get you close is to turn it all the way in (gently!) and then back it out 1-1/2 turns.
Then set the idle speed up as high as you need to in order to get it to keep running. Adjust the idle mixture for best idle (highest speed, smooth). The drop the idle speed as low as you can and have it keep running and repeat.
When you are all done, take it for a hard ride and then repeat the process.
