01 CR250 Rear Shock Adjustment

When adjusting the rear shock, does the punch mark on the rebound adjuster and low speed damping adjuster for compression, HAVE to align with the punch marks on the shock absorber, just like the high speed damping adjuster?

If so, I guess I have to adjust them by one full turn everytime?

No, put it at any "click" or position you want. I believe one full turn would be 4 clicks, which in some cases could make quite a difference...
