I have a DT 100 Enduro and currently I have a tire on the rim that is size 3.00 -18, which is a dual-sport tire. I have a spare tire that is an off-road tire BUT the size is just a little bit bigger, which is 3.50 - 18. My question is will this tire fit onto my rim since it is only a little bit bigger? And would I be able to use the same tube?

It'll probably fit the rim, and the tube shouldn't be a problem they tend to cover a range of sizes anyway. But how is your clearance at the chain and swingarm? You can expect it to stick out around 1/4 inch at either side. It will likely be a little taller as well. You should also expect a little different handling with it. The rear will sway more.

FWIW, my first bike was a '73 Suzuki TC100. Stock tire was a 3.00 x 18 trials type, but a 3.50 knobby fit just fine. No tube, rim, chain or swingarm problems.
