Now that you have the pilot sorted and an easy adjust fuel screw it shouldn't be hard to tune it.

To a certain extent, yes, you can get similar results with a step up or down on the pilot. The difference will be the range of tuning you can do. You don't want to run the 42 at 2 turns out in the summer then try to run it in the winter. I jetted mine to run the fuel screw a bit rich when it's cold so I don't have to change it in the spring/summer. It ran fine in 85 degrees at about 1.25 turns.

The main will be the same as the pilot, larger jet for colder temps. Keep going richer until you feel the power peak out in the top end. Unless you want to start chopping apart expensive spark plugs, the seat of the pants dyno works fine for tuning the main.
